Ultraviolet Sensitive Factors in the Cytoplasm That Affect the Differentiation of Euglena Plastids

چکیده
The bleaching phenomenon in Euglena gracilis strain z offers a unique possibility for investigating the hereditary nature of plastids. Trea tment of Euglena cultures with streptomycin, elevated temperature, or irradiation with a mild close of U V light causes the permanent loss of the capacity to develop chloroplasts in the growing cultures. Early explanations of this phenomenon were that the bleaching treatment differentially inhibited the growth of the plastids or killed them, thus resulting in cells which lacked plastids. In effect the bleaching treatment presumably "cured" the Euglena ceils of their plastids. Recently we reported (1) that the plastids of the bleached Euglena cells did not disappear. The presence of proplastids within cells of bleached Euglena was demonstrated by phase and fluorescence microscopy as well as by tests of the biochemical properties which are usually associated with plastids. 1 It was concluded that the changes induced in Euglena in bleaching are of the nature of mutations affecting the plastids rather than the entire loss of these structures. The bleached plastids are to be regarded as proplastids that can no longer differentiate to chloroplasts. In a quanti tat ive study of the action of U V light

Ultrastructural changes in Euglena after ultraviolet irradiation.
The structural changes associated with the ultraviolet-induced bleaching of light-grown cells of Euglena gracilis were investigated. Our lightand electron-microscopic observations of the bleaching process indicate that there is a continuity of plastid structure in cells 5 generations after receiving a bleaching dose of ultraviolet light. There seems to be a continuous dilution of the plastid th...
متن کاملA Cytoplasmic Regulatory Mutant of Euglena: Constitutivity for the Light-Inducible Chloroplast Transfer RNAs.
An ultraviolet-induced cytoplasmic mutant (G(1)BU) of Euglena gracilis Klebs var. bacillaris Pringsheim is described. G(1)BU, in addition to being golden in color and containing smaller amounts of chlorophyll than wild type when grown in the light, is a regulatory constitutive mutant for the light-inducible chloroplast isoleucine and methionine tRNAs; i.e., these two tRNAs are present in dark-g...

Euglena gracilis offers a particularly suitable system for investigating some of the fundamental processes involved in the provision, transmission, and translation of information required for the building of specific cytoplasmic structures. Cells grown in the dark do not have a defined chloroplast structure; they are etiolated cells (I). Light induces profound changes in the morphology and phys...

Four mutant strains of Euglena gracilis have been isolated after treatment of wild type cells with ultraviolet light or the chemical mutagen nitrosoguanidine. None of the mutants is capable of autotrophic growth or photosynthetic carbon dioxide fixation. The mutant strains contain normal amounts of the enzymes of the reductive pentose phosphate cycle and are qualitatively similar to the wild ty...
